The inaugural event was organised as a major fundraiser for the Creek to Creek group and will consist of a kayak paddle, run and mountain bike ride.
Creek to Creek is a community group where volunteers have banded together to restore and renovate the Queens Lake trail.
Creek to Creek president and organiser of the challenge, Peter Nash said competitors will enjoy the off-road event, which features areas of national park, state forest and crown land.
"If you love the outdoors, have a genuine passion for fun and adventure and want to push yourself to new limits then this is the event for you," he said.
People can enter the event as individuals or participate in a relay team of three members.
Registration for the event closes on Thursday, March 28.
The Queens Lake trail runs from the Lakewood shopping complex along the foreshore and through Henry Kendall Reserve to West Haven. The group hopes to extend the trail to Herons Creek and Stingray Creek Bridge.
Recently the group unveiled a new bench located between Laurieton Lakeside Aged Care and the Queens Lake Retirement Village at West Haven.
Peter said the structure would encourage social interaction and allow all ages to enjoy the natural environment.
In the future, the Creek to Creek group plans to install covers for the benches so that they can provide shelter for people from the weather elements.
