Blood service spokesperson Nicky Breen says the current cold and flu season has hit the centre hard with a number of cancellations putting pressure on blood supplies.
The call out includes people who may have given blood in the past or current donors.
Ms Breen said the current cold and flu season was one of the worst on record.
"Our data is showing that up to 1400 donors each day are cancelling their appointments across Australia," Ms Breen said.
"We haven't seen cancellations this high since March 2017, and as the number of people suffering cold and flu symptoms increases, it limits the number of regular donors who are able to give.
"Last week, we called for an additional 5900 people to give blood in the run up to the Queen's Birthday long weekend and the response has been phenomenal.
"However, this year's cold and flu season has hit hard so we are continuing to urge people to donate blood over the coming months."
The Port Macquarie Donor Centre needs an extra 81 donors to roll up their sleeves and donate blood between now and the end of the next week.
"We need people who are fully recovered from colds or the flu, and those who have avoided it, to take the place of those who are unable to answer our call," Ms Breen added.
On Wednesday, Kinchela's Learne Lincoln was making her 25th donation while fellow Kempsey resident Nicky Thompson was dressed as Superwoman in a bid to help draw attention to the urgent need for blood donations.
"I am promoting giving blood through my business Facebook page and hopefully more people come in and give blood," Ms Thompson said.
Historically, cold and flu season can place a strain on the blood service as symptoms prevent existing donors from making their life-saving donations.
People are still able to donate if they have had a flu vaccination and donors affected by cold and flu symptoms are able to give blood seven days after making a full recovery.
